Abstract
Tumor suppressor genes are vital for preventing cancer by controlling cell growth and repair. Their loss or inactivation through mutation or epigenetic silencing is common in cancer. This review discusses the roles of key tumor suppressors like RB, TP53, BRCA1/2, PTEN, and APC in cell cycle regulation, DNA repair, and signaling pathways. Genomic instability and epigenetic alterations are highlighted as significant mechanisms of their inactivation. Emerging therapeutic strategies aim to restore tumor suppressor function for cancer treatment.
